Characterization and Purification of a Nuclear Factor that Binds to an Upstream Regulatory Element of the Rat Apolipoprotein E Gene

Abstract
We have identified and characterized a nuclear protein that binds to an upstream regulatory element(URE-) 3 which corresponds to the nucleotides -101 to -81in the 5'-flanking region of the rat apolipoprotein(apo-) E gene. Results from gel
mobility
shift
assays showed that there are similarities and differences between the human URE3 bindng protein(HURE3P) and rat URE3 binding protein (RURE3BP) with respect to the sensitivity to metal ions, pH, and competition against various transcription
regulatory
elements including GC box, AP-1, FSE-2 binding sequences. To characterize RURE3BP further, we have purified RURE3BP from rat liver nuclear extracts using RURE3 oligonucleotide-specific affinity column following gel filtration column
chromatography.
The
relative molecular mass of the purified RURE3BP was 98,800 dalton and it showed high specific binding activity for RURE3 probe.
